FGD5 is identified as a metabolic vulnerability that increases PDAC sensitivity to the glycolysis inhibitor 2-deoxy-D-glucose (2-DG) and a mechanistic framework supporting combinatorial strategies that couple glycolysis inhibition with perturbation of FGD5–PGK1-associated proteostasis is provided.
Findings uncover an OTUD4-FXR1-HIF1A signaling axis that promotes glycolysis, which in turn functionally drives gastric cancer progression, and thereby provides a potential prognostic biomarker and therapeutic target.

Ovarian cancer (OV) remains one of the most lethal gynecological malignancies, with limited effective early detection strategies and therapeutic options. Despite advances in platinum-based chemotherapy and targeted therapies, including PARP inhibitors and immune checkpoint inhibitors, treatment failure and recurrence r...
A metazoan-conserved dual degradation inhibition cascade whereby energy stress coordinates acetylation-phosphorylation crosstalk that simultaneously enhances glycolytic output and suppresses two protein degradation systems is reported.

ABSTRACT Metabolic reprogramming is a hallmark of gastric cancer (GC); however, the role of transcriptional regulators in modulating glycolysis remains incompletely understood. By integrating proteomic, transcriptomic, and CRISPR‐based functional screening datasets, we identified WDR12, a WD40‐family protein that is up...
