Abstract

Citrinin (CTN), a nephrotoxic and hepatotoxic mycotoxin frequently found in red yeast rice, requires rapid and reliable detection. Here, six CTN haptens were rationally designed using computational chemistry, and two optimal structures were selected for generating CTN-specific aptamers via systematic evolution of ligands by exponential enrichment (SELEX). Based on these aptamers (Apts), four analytical platforms were developed, including enzyme-linked aptamer sorbent assay (ELASA), aptamer-based lateral flow strip (Apt-LFS), hybrid enzyme-linked aptamer-antibody assay (ELAAA), and hybrid lateral flow strip (Hybrid-LFS). The limits of detection were 1.9, 500, 10, and 250 ng/mL, respectively. In spiked red yeast rice samples, ELASA and ELAAA showed average recoveries of 102% and 104%. Analysis of 15 commercial samples showed excellent consistency with HPLC, while Apt-LFS and Hybrid-LFS identified four positive samples according to Taiwan Food and Drug Administration (TFDA) limits. This study provides a sensitive and practical strategy for CTN monitoring in complex food matrices.

From DNA Aptamer Selection to Label-Free Fluorescent Biosensor for Chebulinic Acid

Chebulinic acid is a bioactive hydrolyzable tannin of increasing interest in natural product research, quality control, and pharmaceutical studies. In this work, four candidate DNA aptamers were identified using a restriction endonuclease-assisted SELEX (REase-SELEX) strategy, and Aptamer 1 was selected based on it...

A Dual-T-Line Immunochromatographic Assay Based on Computational Chemistry-Guided Hapten Design for Rapid On-Site Detection of Chloroneb in Agricultural and Environmental Samples

The health risks posed by chloroneb (CHL), a representative fungicide, in agricultural products and the environment necessitate rapid detection methods. Here, computational chemistry-aided rational hapten design yielded a high-performance monoclonal antibody against CHL (IC50 = 2.63 ng/mL), guided by analysis of its...

From Cross-Reactivity to Specificity: A SELEX-Derived Aptamer Sensor Array for Aristolochic Acid I and Aristolactam I

Aptamer selection often yields multiple sequence families that recognize different epitopes or bind through distinct molecular interactions. Rather than using a single aptamer for detection, we demonstrate that these complementary aptamers can be integrated into a cross-reactive sensor array to enhance molecular disc...

Serum-compatible screening and truncation optimization of copeptin aptamers for TdT-amplified fluorescent detection.

Copeptin is a clinically relevant biomarker for early diagnosis and prognostic assessment of acute myocardial infarction (AMI). Traditional antibody-based assays suffer from long testing time, high cost and complicated operation. Herein, we first screened high-affinity copeptin aptamers under serum-mimicking conditions...

Rapid Detection of Dextromethorphan in Saliva and E-Juice Based on Highly Selective Aptamers

Dextromethorphan (DXM) is a widely used cough suppressant, and its escalating abuse is a pressing global health concern that necessitates rapid screening tools. However, rapid methods for detecting DXM in complex matrices remain highly limited. Here, we report the first isolation and engineering of DNA aptamers speci...
