Nuclear IDH3A Drives Transcriptional Programs in Melanoma via the YBX1–JUN/FOS Axis

It is demonstrated that IDH3A is significantly overexpressed in melanoma, primarily due to DNA copy number amplification, and a noncanonical role of IDH3A is uncovered as a nuclear regulator of transcription via YBX1, offering novel insight into metabolic enzyme reprogramming in melanoma.

Epigenetically driven GAS2L3 enforces non-canonical cell cycle progression to promote osimertinib resistance in lung adenocarcinoma

These results uncover a metabolism-epigenetics–cell cycle interface that may represent a therapeutic vulnerability in EGFR-mutant LUAD and identify a glycolysis–H4K8la–GAS2L3 axis that drives noncanonical cell-cycle reprogramming independently of classical Cyclin–CDK activation, thereby promoting acquired osimertinib r...

NAT10 promotes prostate cancer progression by acetylating mRNA of MARK2 and activating mTOR/HIF-1α/C-Myc pathway to enhance glycolytic reprogramming.

Mechanistic investigations demonstrated that NAT10 elevates MARK2 expression, activating the mTOR/HIF-1α/C-Myc signaling cascade, thereby promoting aerobic glycolysis in PCa cells, suggesting that targeting ac4C mRNA modifications may provide a novel therapeutic strategy for managing prostate cancer.

METTL15-mediated m4C modification regulates glucose metabolic reprogramming via c-Myc-dependent mechanisms and promotes hepatocellular carcinoma progression.

Although aerobic glycolysis is a hallmark of hepatocellular carcinoma (HCC) progression, the underlying epitranscriptomic mechanisms remain poorly understood. Here, we identify METTL15, an N4-methylcytidine (m4C) RNA methyltransferase, as a novel oncogenic driver of this metabolic reprogramming. METTL15 is significantl...

Targeting FEN1 disrupts EIF4A3-mediated MUC1-C autoregulation: a novel strategy to suppress lung adenocarcinoma progression

The FEN1/EIF4A3/MUC1-C axis is established as a novel mechanism driving LUAD progression by concurrently regulating intrinsic malignancy and extrinsic immune evasion, presenting FEN1 targeting as a promising dual-hit therapeutic strategy.
