TL;DR

Functional analyses demonstrated that KH2-KH3 linker and KH3 domain of PCBP, plays an important role in the AsPCBP-ferritin interaction and that deletion of these regions, unexpectedly increased ferritin binding affinity but impaired iron delivery, indicating that these regions are dispensable for complex formation but essential for efficient iron transfer.

PEDV envelope protein promotes viral replication by remodeling host iron homeostasis via the TRIM28-KLF15-FPN axis

ABSTRACT Porcine epidemic diarrhea virus (PEDV), a member of the Alphacoronavirus genus, causes severe enteric disease with up to 100% mortality in suckling piglets. Iron is an essential micronutrient for the replication of various pathogens, including viruses, which hijack host iron resources to support their prolifer...

Acyl carrier protein is essential for apicoplast biogenesis in malaria parasites independent of fatty acid synthesis

Acyl carrier protein (ACP) and its 4-phosphopantetheine prosthetic group canonically function as the soluble scaffold for acyl chain assembly and elongation during type II fatty acid biosynthesis (FASII). Plasmodium malaria parasites retain a FASII pathway in the apicoplast organelle that has been the subject of consid...

Biophysical and molecular determinants of NCOA4-mediated ferritinophagy at the ferroptosis-inflammation interface: Compartmental iron transport, lipid oxidation, and causal constraints.

Ferritinophagy links molecular recognition of ferritin to the redistribution of redox-active iron across intracellular compartments. This critical narrative review examines nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy as a multiscale process in which NCOA4-FTH1 binding and ferritin condensation govern...

The Plasmodium heme detoxification protein functions in mitochondrial protein synthesis

Malaria blood-stage parasites digest ~80% of host cell hemoglobin within a degradative vacuole, releasing heme that is detoxified by sequestration into hemozoin crystals. Although essential for survival and a validated drug target, the mechanisms of heme biomineralization remain unclear. Here, we study the parasite’s H...

Conserved copper(I)-binding auxiliary domains link virulence-associated LPMOs to copper homeostasis in the intestinal environment

It is demonstrated that these multidomain LPMOs from food-borne intestinal disease-causing Gram-positive bacteria from the genera Bacillus and Listeria contain conserved, yet structurally distinct copper(I)-binding motifs on their non-catalytic third domain.

Promiscuous Metal Site in Hepatitis B Virus X Protein Binds an Fe-S Cluster.

Fe-S clusters are emerging as important cofactors in viral replication but are frequently misassigned as Zn due to their O2-sensitivity and overlapping cysteine-based coordination chemistry. The Hepatitis B virus regulatory protein HBx, which is essential for viral replication and hepatocarcinogenesis, has long remaine...
