Multiomic and Spatial Profiling of Colorectal Tissue Reveals Viral Persistence and Immune Dysregulation in Long COVID

Biology

TL;DR

A tissue-based transcriptional environment in which SARS-CoV-2 activates innate myeloid immune signaling, driving chronic inflammation while simultaneously downregulating pathways responsible for immune-mediated clearance of infected cells is identified, including antigen presentation, phagocytosis, cytotoxic immune cell trafficking, and granzyme production.

Gut microbiome signatures during acute infection are associated with long COVID

Using machine learning, it was showed that the gut microbiome carried a modest signal for subsequent LC, but model performance was insufficient for clinical prediction, likely reflecting the heterogeneous nature of LC.

Progressive Post-Acute Gut Microbiome Disruption Following SARS-CoV-2 Infection in Mice

SARS-CoV-2 infection is increasingly recognized to produce long-lasting physiological disturbances that extend beyond the acute phase. The gut microbiome has emerged as a potential contributor to post-infection outcomes, yet the longitudinal progression of microbial disruption following infection remains poorly defined...

Metabolic, viral, and immune phenotypes in long COVID

Context Long COVID is characterized by persistent symptoms ≥ 3 months after acute SARS-CoV-2 infection. To date, the underlying pathophysiology is unclear. Objective To characterize the immune and metabolic features of Long COVID and the potential role of viral persistence in adipose tissue. Design Case-control, cross-...

Exploratory Profiling of Circulating microRNAs (miRNAs) in Patients with Post-COVID-19 Syndrome

Post-COVID-19 syndrome (PC) is defined by the persistence of symptoms over 12 weeks after infection with SARS-CoV-2, without any other diagnosis. These symptoms can affect multiple systems with neurological, hemodynamic, and respiratory disorders. Exacerbated activation of the innate immune response mediated by cytokin...

Case Report: Post-mortem analysis of long-term inflammation after mild COVID-19

Persistent inflammation and tissue injury may contribute to post-acute COVID-19 syndrome (PACS). However, direct tissue evidence from mildly-infected individuals sampled long after recovery remains exceptionally limited. Here, we performed post-mortem bulk RNA-seq and GeoMx digital spatial profiling of lung, heart, and...

NINJ1 Defines a Hyperinflammatory Monocyte State Associated with Severe COVID-19

Background/Objectives: Severe coronavirus disease 2019 (COVID-19) is characterized by dysregulated immune activation and excessive inflammation; however, the specific immune cell states that drive disease progression remain incompletely understood. Methods: Here, we integrated single-cell RNA sequencing analysis of per...
