CHAPTER 4 The HSPG binding domain of APRIL serves as a platform for ligand multimersation and cross-linking.

TL;DR

It is shown that the lack of HSPG interaction can be compensated by artificial cross-linking, which indicates that the HSPG-binding domain in APRIL is not necessary to allow for signalling through HSPG, but instead mediates ligand cross-linking and oligomerisation.

Design of a high-affinity IL-21 decoy receptor as a novel therapeutic strategy for autoimmune disorders.

Elevated levels of the pro-inflammatory cytokine Interleukin 21 (IL-21) contribute to the progression of autoimmune and inflammatory diseases by binding to its receptor (IL-21R) and activating detrimental signaling pathways in immune cells. Consequently, we developed a recombinant decoy receptor protein designed to com...

Siglec-1-targeting single domain antibodies block or enhance pathogen binding to dendritic cells.

Siglec-1 (Sialoadhesin/CD169), expressed on myeloid cells, is a receptor for sialic acids that mediates adhesion to host cells and pathogens. However, certain pathogens exploit Siglec-1 binding for further dissemination and interfering with this binding may serve as potential therapeutic interventions. Here we identifi...

Structural basis for tetraspanin-dependent surface export and adhesive function of integrin α3β1

Integrin α3β1 (ITGα3β1) is a member of an integrin subfamily that binds to laminin proteins and promotes attachment of epithelial cells to the basement membrane. ITGα3β1 forms a complex with the tetraspanin CD151, and loss-of-function mutations in both ITGα3 and CD151 cause epidermolysis bullosa, a severe skin blisteri...

Characterization of glycosaminoglycan interactions with the chemokine CCL20 (MIP3α) and its carboxyterminal antimicrobial and antibiofilm peptide.

The chemokine CCL20/MIP3α plays an important role in the migration of dendritic cells and several lymphocytes. This 70-residue protein is also implicated in various diseases, such as rheumatoid arthritis, psoriasis, sepsis and numerous forms of cancer. In addition, as the most basic human chemokine, CCL20 also displays...

Synergistic Activity of APOL3 and GBP1 in Antigen Cross-Presentation: A Model

It is proposed that phagolysosomal membrane disorganization by GBP1 allows pore formation by APOL3, through transmembrane insertion and cardiolipin solubilization in the inner coating of phagolysosomal membrane megapores.
