Highlights • Single-cell transcriptomic profiling of multicenter pancreatic cancer radiotherapy cohorts identified PLPP2 as a key driver of radiotherapy resistance and immune microenvironment remodeling.• Random forest model identified PLPP2 as an important marker for promoting malignant progression, immune dysregulation, and poor clinical outcomes in pancreatic cancer.• PLPP2 is potential to be a marker for predicting metabolic status, immune landscape, and response to immune checkpoint inhibitors in patients with pancreatic cancer.
