Abstract

Producing enantiopure meso-2,3-butanediol (meso-2,3-BDO) with minimal organic acid byproducts is critical for its commercial applications in specialized polymers and cosmetics. In this study, we developed a highly selective and robust Klebsiella oxytoca platform using stepwise metabolic engineering and bioprocess optimization. Starting with a lactate- and formate-deficient parent strain (Δldh/ΔpflB), we sequentially deleted poxB and frdBC to suppress acetate and succinate byproducts. To resolve recalcitrant succinate accumulation caused by NADH surplus during micro-aerobic fed-batch fermentation, we implemented dynamic redox control strategies (nutrient supplementation and agitation shifts) and permanently sealed the reductive TCA branch by deleting mdh. Furthermore, to achieve ultra-high stereoisomeric purity, we disrupted the isomer interconversion network by knocking out dar and gldA. The resulting engineered strain (GSC-P8) was cultivated using the optimized redox-control bioprocess. This integrated scale-up fermentation completely abolished undesired stereoisomers, strictly maintaining meso-2,3-BDO selectivity at approximately 97.9%. We achieved a prominent meso-2,3-BDO titer of 125.2g/L and a volumetric productivity of 2.84g/L/h, while restricting each acidic byproduct to below 2.0g/L. This study demonstrates a commercially viable strategy for pure meso-2,3-BDO production through rigorous intracellular redox homeostasis.

Engineering Klebsiella oxytoca for enhanced 2,3-butanediol production from gluconate

Historically, microbial production of 2,3-BD has relied predominantly on glucose as the substrate. However, its fermentation necessitates microaerobic conditions due to inherent redox imbalances. This limitation complicates process control and can lead to undesirable late-stage consumption of 2,3-BD. This study...

Highly efficient production of 1,3-propanediol by CRISPR-Cas12a-edited Klebsiella pneumoniae using glycerol and lignocellulosic hydrolysate as co-substrates.

Efficient biosynthesis of 1,3-PDO, a key bio-based chemical, depends on precise regulation of the host metabolic network. In this study, a heterologous CRISPR-Cas12a genome editing system was established and systematically optimized in Klebsiella pneumoniae, enabling efficient and stable genome editing (75-100% efficie...

Engineering of central metabolism for high-yield 1,3-propanediol production using co-substrates by Klebsiella pneumoniae.

1,3-Propanediol is a versatile C3 diol serving as a key building block used in various industries. Klebsiella pneumoniae represents an attractive microbial chassis for 1,3-PDO production due to its endogenous 1,3-PDO and Vitamin B₁₂ biosynthetic pathways. However, low carbon flux and by-product formation remain bottlen...

Biosynthesis of 4-hydroxyvalerate from glycerol in engineered Pseudomonas putida KT2440 and its application for the production of Poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-4-hydroxy valerate) (P(3HB-co-3HV-co-4HV)) terpolymer.

The 4-hydroxyvalerate (4HV) is a key intermediate that serves as a precursor for γ-valerolactone (GVL) and terpolymers, contributing to the synthesis of liquid fuels and biodegradable plastics with improved properties. While 4HV is typically derived from levulinic acid (LA) via chemical catalysis, these methods are lim...
