Three amino-terminal variants identified in hypoglycemic patients underlie the molecular basis for the clinical variability of AADC deficiency, providing a molecular basis for the clinical variability of AADC deficiency.

Abstract

AADC deficiency is a rare autosomal recessive disorder characterized by motor/neurodevelopmental symptoms. Approximately one-third of patients experience recurrent hypoglycemia, with or without neurological involvement. To investigate whether specific variants identified in hypoglycemic patients underlie this metabolic phenotype, we characterized three amino-terminal variants. The crystal structure of p.Asp15Gly revealed disruption of an inter-subunit electrostatic network stabilizing the enzyme interface. Computational analyses predicted that p.Val33Leu and p.Gly36Arg perturb an essential catalytic element. Functional studies showed that p.Asp15Gly retained substantial catalytic activity and coenzyme affinity despite reduced stability, whereas p.Val33Leu and p.Gly36Arg exhibited increasing structural destabilization, reduced coenzyme affinity, and severe catalytic defects. These findings reveal distinct functional roles of the amino-terminal-domain, providing a molecular basis for the clinical variability of AADC deficiency.

Structural and biochemical characterization of disease-associated mutations in the human mitochondrial processing peptidase α-subunit.

The mitochondrial processing peptidase (MPP) is an essential heterodimeric protease responsible for cleavage of N-terminal mitochondrial targeting sequences (N-MTSs) from most nuclear-encoded mitochondrial proteins following import into the mitochondrial matrix. Pathogenic variants in the α-subunit of MPP (PMPCA) cause...

Integrative Evolutionary and Structural Analysis of GBA1 Provides Insights Into Mutation-Specific Effects Associated With Parkinson's Disease.

GBA1 encodes lysosomal acid β-glucosidase (glucocerebrosidase [GCase]), a key lysosomal enzyme and one of the most important genetic factors in Parkinson's disease (PD). Yet how different GBA1 variants influence PD-related molecular pathology remains incompletely understood. Here, we integrated phylogenetic, comparativ...

Disease-associated amino acid substitutions outside canonical motor motifs impair Myo1e localization and activity

Steroid-resistant nephrotic syndrome (SRNS) is a severe kidney disorder characterized by proteinuria and disruption of glomerular filtration, often driven by genetic defects in genes encoding proteins required for podocyte functions. Among these, MYO1E, encoding the actin- and membrane-associated motor protein Myosin 1...

Exploring the Role of Positive Selective Sites in Enhancing Structural Stability and Functional Efficiency of Human Lipase A.

Lysosomal acid lipase (LAL), encoded by the LIPA gene, is essential for lipid metabolism, and its deficiency causes Wolman disease and cholesteryl ester storage disease. Understanding the structural determinants of LAL stability and function is crucial for protein engineering and therapeutic development. We integrated...
