Prolyl endoprotease (An-PEP) for hydrogen-deuterium exchange mass spectrometry and protein structural studies

· 0 citations · 6 references

TL;DR

Due to its Pro-preference and resistance to low pH, An-PEP is an archetype enzyme for HDX-MS, and higher sequence resolution could be achieved, especially for Pro-containing protein regions in the alpha subunit of Hb, revealing new protected Hb regions that were not observed with pepsin.

LemonCatcher Acidic Pull-Down Enables Selective In-Cell Hydrogen-Deuterium Exchange Mass Spectrometry

LemonCatcher is created, a protein superglue that spontaneously forms an amide bond to the LemonTag peptide under these harsh acidic and cold quench conditions, even at -20 °C, and SelQueX makes possible selective characterization of protein structural dynamics within the complex cellular milieu.

SIMPATICO - Streamlined isolation of the N-terminome via phosphonate tagging and coordination-based depletion.

Protein processing is critical for regulating cellular functions. Therefore, there is considerable interest in developing effective proteome-wide strategies to identify protease cleavage products to enhance our understanding of proteolytic pathways and their perturbation in diseases. Here, we present a streamlined N-te...

Optimized detection of acidic ADP-ribosylation by PARP14 in vitro using multi-modal mass spectrometry reveals putative autoregulatory patches.

ADP-ribosylation is a highly dynamic process that is increasingly linked to a diverse set of cellular functions and pathophysiological outcomes. The modification of target substrates (e.g., proteins, DNA, RNA) involves ligation to an array of different chemical moieties. Recent work has shown that the ester bonds forme...

Mismetallation reveals metal tolerance and enhanced stability of human deoxyhypusine hydroxylase.

Hypusination is a unique post-translational modification found exclusively in eukaryotic translation factor 5A (eIF5A). In this reaction, a specific lysine residue in eIF5A is converted into hypusine residue. The modification comprises two enzymatic steps: a NAD-dependent deoxyhypusine synthase (DHS) transfers the 4-am...

Dynamics of the Myoglobin/Heme Complex: A Limited Proteolysis Perspective

The dynamics and stabilities (i.e., physicochemical properties) of proteins and protein complexes are dictated by the solution environment. Changes in the solution environment may alter the distribution of states present in solution (i.e., the free-energy landscape) and thus their physicochemical properties. This wor...

Enzymatic formation of a conserved isoaspartate in ribosomal protein uS11

Isoaspartate (isoAsp) formation is typically viewed as a “molecular clock” through nonenzymatic degradation of aspartate or asparagine during protein aging. Here we report a nearly universal enzymatic pathway for the formation of a conserved isoAsp in the bacterial ribosomal protein uS11. Proteome-wide protein-protein...
