TL;DR

This study shows that the conserved capsule gene CpsA is not strictly required for capsule attachment but instead works together with other LytRIZCpsAIZPsr (LCP) ligases, however, it requires the essential WalRK signaling system to maintain cell envelope integrity.

Protein–Protein Interactions Between Peptidoglycan, Lipopolysaccharide, and Phospholipid Biosynthesis Enzymes in Escherichia coli

Gram‐negative bacteria are protected by a three‐layered cell envelope and must tightly coordinate the biosynthesis of phospholipids (PL), peptidoglycan (PG) and lipopolysaccharides (LPS) to maintain envelope integrity. In Escherichia coli, the inner membrane protein LapB (YciM) plays a critical dual role in LPS homeost...

A Mobile Glycosylation Locus Modulates Cell Wall Architecture in Lactobacillus crispatus.

Lactobacillus crispatus dominance in the vaginal microbiome is associated with beneficial health outcomes, yet strain-level variation and its implications remain poorly understood. Here, we resolve the genomic context of three glycosyltransferase gene fragments (GT1-3) previously linked with dysbiotic states. Long-read...

FlgJ cell-wall hydrolyzing activity enhances, but is not required, for flagellum assembly in Salmonella enterica

Salmonella can assemble functional flagella, even when the acetylglucosaminidase activity of FlgJ is genetically inactivated, showing that early steps in flagellar assembly are more diverse than previously thought and that differences in cell wall structure between gram-positive and -negative bacteria likely determine...

Structural and Functional Interrogation of Active Streptococcus pneumoniae Sortase A

Sortase A (SrtA) enzymes covalently anchor surface proteins to Gram-positive bacterial cell walls, promoting colonization and virulence. In Streptococcus pneumoniae, previous studies identified both a domain-swapped dimer and an active refolded monomer, but the active enzyme has not been characterized at the structural...
