TL;DR

This study analyzed specific residues within the C-terminal α11 helix that are critical for stable association of Orc6 with the ORC complex in Drosophila and proposed that this NLS drives human protein to the nucleus and compensates for weaker Orc6-ORC(1-5) interactions by increasing the nuclear concentration of Orc6 and shifting the equilibrium toward formation of the fully assembled ORC complex at the DNA.

Replication protein A1 from L. donovani interacts with RECQb via the C-terminal OB fold and stimulates its helicase activity on branched substrates.

The results reveal a new C-terminus-mediated RPA-RecQ interaction in L. donovani that resembles the prokaryotic SSB-RecQ interaction mechanism, while maintaining functional conservation of eukaryotic DNA metabolism.

A microeukaryotic PPR-DYW protein supports multisite C- and A-deamination RNA editing

RNA editing in mitochondria is vital for many eukaryotes. In plants, mitochondrial C-to-U deamination editing is catalyzed by tens to hundreds of PPR-DYW proteins, each typically dedicated to a specific site. Where retained, PPR-DYW family expanded independently in all eukaryotic groups examined here—except in marine m...

The eumetazoan origin of PRDM9 function revealed by cnidarian genome analysis

Where recombination takes place is crucial as it determines the position of genetic reshuffling, which facilitates species evolution and adaptation. In many vertebrates, PRDM9 determines the location of double-strand breaks that initiates meiotic recombination. However, this only applies for Prdm9 orthologs that posses...

A systematic interactome of Saccharomyces cerevisiae SET1C expands its functional landscape and identifies candidate regulatory connections

Set1 is the catalytic subunit of SET1C or COMPASS, which methylates histone H3K4 and serves as a scaffold for the association of seven tightly bound polypeptides. We have employed yeast two-hybrid screenings to determine the interactome of Set1 and each subunit, providing a unique resource for exploring known and novel...

Single-molecule chromatin tracing reveals a diversity of megabase heterochromatin domains.

Chromosomes fold into distinct domains that regulate transcription, replication and repair. In addition to well-characterized topologically associating domains (TADs) and compartments, the diversity of heterochromatin domains remains poorly defined at the sequence level. Here, using single-molecule tracing of nascent h...
