In conclusion, small-molecule-based therapies represent a promising multi-targeted strategy for PD; however, key translational challenges and unresolved questions remain, including optimisation of pharmacokinetics, target specificity, and blood-brain barrier penetration and validation in clinical settings.
This review focuses on translational insights linking α-syn pathology to dysregulated stress-response and protein quality-control pathways, intending to identify potential targets for disease-modifying intervention.

Abstract Ageing is a primary risk factor for neurodegenerative disorders, including Parkinson's disease (PD). As individuals age, their cells become less efficient in maintaining protein homeostasis, leading to an increased likelihood of protein misfolding and aggregation. A hallmark of PD and other synucleinopathies i...

This review synthesises current evidence on how mutations in Leucine-Rich Repeat Kinase 2 (LRRK2), the most common genetic cause of familial PD, fundamentally corrupt glial handling of extracellular α-synuclein and evaluates emerging therapeutic strategies targeting LRRK2 kinase activity, NLRP3 inflammasome activation,...

It is proposed that a systems-level approach can be taken where PD is understood as a disease characterized by the imbalance in proteostasis, and potential treatment modalities include small molecules targeting lysosome function, CMA enhancers, gene therapy, and antibodies against extracellular α-synuclein.

While amyloid-β (Aβ) has historically dominated the research landscape of Alzheimer’s disease (AD), the limited clinical success of Aβ-centric therapies has redirected focus toward tau pathology, which correlates more robustly with cognitive deterioration and synaptic dysfunction. Transcending the traditional linear...
