It is demonstrated that ex vivo expanded autologous HSCs act as a strong biological stimulant, significantly aiding leukocyte proliferation and immune signalling and providing a safe, human-compatible, and effective framework for good improvement of such haematological factors.
A protocol to isolate and expand MSCs from human umbilical cord tissue and characterize their cell surface protein expression by flow cytometry, self-renewal potential by colony-forming assay, and tri-lineage differentiation capacity is outlined.

A scalable culture system to preferentially expand human CD4− iNKT cells directly from total peripheral blood mononuclear cells (PBMCs) using alpha-galactosylceramide (α-GalCer) and optimized cytokine conditions and IL-15 was the most effective cytokine.

Although allogeneic hematopoietic stem cell transplantation has long been the only curative therapy for nonmalignant blood diseases, recent clinical successes have demonstrated the transformative potential of autologous gene therapies as definitive treatments for these disorders. By providing durable life-changing clin...

This chapter describes two in vitro assays designed to assess the hematopoietic stem/progenitor cell (HSPC)-supporting capacity of primed MSCs, and the other is specifically aimed at their ability to support the growth of primitive HSCs.

The hematopoietic system is one of the most rapidly renewing systems in the body. Hundreds of billions of blood cells are produced daily in adults, and maintaining this process depends on a small population of hematopoietic stem cells (HSCs). HSCs, which support adult hematopoiesis, are formed in the fetal liver and th...
