Abstract
Decalin-forming Diels–Alderases (pericyclases) exemplify how Nature repeatedly recruits unrelated protein folds to control [4 + 2] cycloadditions, yet exo-selective intramolecular Diels–Alder (IMDA) reactions that yield cis-fused decalins remain rare and poorly understood. Here, we uncover a previously unrecognized bacterial pericyclase family that enforces an exo-IMDA trajectory in polyketide assembly. Using alchivemycin A, a macrocyclic antibacterial polyketide whose scaffold is defined by a highly functionalized cis-decalin core, we identify AvmG as the dedicated decalin-forming enzyme. Disruption of avmG diverts pathway flux to trans-decalin shunt isomers consistent with spontaneous, endo-biased cyclization that cannot be processed by the downstream macrocyclase, whereas AvmG enables formation of the authentic cis-decalin intermediate. AvmG is unrelated to known [4 + 2] cyclases and adopts an integrin α N-terminal domain-like β-propeller architecture, revealing an unexpected scaffold for pericyclic stereocontrol. Genome mining shows that AvmG-like proteins are widespread and enriched in actinomycete polyketide gene clusters; characterization of the lydicamycin homolog LymR establishes conserved exo-IMDA activity in a distinct pathway. Structural and mutational analyses implicate a loop-defined catalytic cavity in steering the kinetically demanding cis-decalin outcome. Together, these findings expand the fold and mechanistic diversity of Diels–Alderases, define an exo-selective β-propeller pericyclase superfamily, and provide a new handle for mining and bioengineering cis-decalin biosynthetic pathways.
Unspecific peroxygenases (UPOs) catalyze the H2O2-dependent monooxygenation of organic substrates with regioselectivity and chemoselectivity; however, their application to sterically demanding, non-natural terpene skeletons has remained largely unexplored to date. Here, we present a computationally-guided, multiparam...

Terpene cyclases (TCs) typically transform acyclic oligoprenyl pyrophosphates through carbocationic cyclization cascades. However, recently discovered bacterial pathways generate compact, cyclized and stereochemically defined noncanonical intermediates through methyltransferases prior to TC catalysis, raising the que...

Maleimide is a rare but powerful pharmacophore in natural products, yet its biosynthetic logic remains poorly understood. Here, we elucidate the biosynthesis of ligiamycins, bacterial decalin–amino-maleimide metabolites from Streptomyces sp. NA07271. Genetic and biochemical studies revealed an unusual dispersed PKS–N...

Pentacyclic triterpenoids (PTs) feature a steroid-like privileged scaffold, but late‑stage functionalization of their inert B‑ring C(sp3)─H bonds remains a formidable synthetic challenge due to the absence of directing groups and the consequent poor regio- and stereoselectivity. Here we report a semi-rationally enginee...
A scaffold-hopping strategy in which a cyanosulfenylcarbene selectively cleaves furan cores, generating dienone intermediates bearing an intrinsic thioaryl leaving group, and provides a conceptually distinct approach to the controlled remodeling of heterocycles is reported.

Euphorbia diterpenoids are a structurally diverse class of natural products renowned for their potent biological activities. Inspired by biosynthetic 1,2-migrations, yet deliberately departing from these pathways, we developed a synthetic strategy that provides collective access to multiple distinct skeletal families t...
