Conformational activation of GSK3β by an environmental toxicant suppresses hedgehog signalling and disrupts ciliary homeostasis

TL;DR

It is shown that endosulfan, a banned organochlorine pesticide linked to congenital and reproductive defects, suppresses hedgehog signalling by driving proteolytic processing of GLI transcription factors into repressor forms, establishing a chemical–biological axis through which an environmental toxicant hijacks core kinase signalling to phenocopy a genetic ciliopathy.

Primary cilia: master conductors of cellular communication in development and disease.

Emerging evidence indicates that cell-type-specific and temporally dynamic ciliary signatures determine signalling competence and disease susceptibility, and defining these signatures and the mechanisms that regulate their plasticity could lead to new therapeutic opportunities for ciliopathies and other cilia-associate...

LRRK2: Molecular Mechanisms in Parkinson’s Disease

Structural, biochemical, and cellular evidence is integrated to advance a unifying hypothesis that PD-associated LRRK2 dysfunction arises from maladaptive stabilization of specific conformational and spatial states within a membrane-responsive signalling network, leading to persistent misregulation of Rab-dependent tra...

A CK2α–G3BP1 signaling axis regulates local translation in developing neurons and is disrupted in OCNDS

Findings establish OCNDS as a disorder of compartment-specific translational dysregulation driven by impaired CK2α–G3BP1 control of RNA granule homeostasis, and establish G3bp1 knockdown rescues translational and morphological phenotypes across all OCNDS alleles.

ZAK kinase at the crossroads of cellular stress: functional plasticity, human pathobiology, and precision therapeutics

Mitogen-activated protein kinase kinase 20 (MAP3K20/ZAK) has emerged as a critical biophysical sensor, uniquely poised at the intersection of environmental stress and cellular fate. Its versatile signaling is primarily orchestrated by two structurally distinct splice variants: ZAKα, which drives the ribotoxic stress re...

Tiny Messengers, Actionable Targets: sEV-Driven Epigenetic Changes in Cancer

Small extracellular vesicles (sEVs) are pivotal mediators of intercellular epigenetic communication in cancer. Following MISEV2023, we use the size- and isolation-based term sEV throughout, because most primary studies cited here cannot resolve which biogenetic route generated the vesicles they analyzed. By selectively...

Canonical and non-canonical STAT3 pathways in epilepsy: Mechanistic insights and translational challenges.

Epileptogenesis is a multifactorial cascade involving neuroinflammation, glial activation, neuronal hyperexcitability, abnormal synaptic plasticity, and cell death. Understanding the epileptogenic changes is critical for improving therapeutic outcomes. Despite extensive knowledge of inflammatory pathways in epilepsy, t...
