Abstract

This study summarizes the automated, transition-metal-free radiosynthesis approach for producing [11C]LCM and its bioisosteric analogue [18F]FLCM, along with preliminary pharmacological and preclinical evaluations of these radiotracers can act as potent brain imaging radiotracer candidates. Radiosynthesis was carried out using an automated synthesis module targeting the terminal 3-oxy site of lacosamide. Physicochemical properties, including hydrophilic behavior and plasma-protein binding profile, were evaluated. Translational validation was performed via in vivo PET/CT imaging and ex vivo biodistribution in healthy Wistar rats. The synthesis module yielded a radiotracer with high radiochemical yield and radiochemical purity. Specific activity was determined to be ~21 .7GBq/μmol for [11C]LCM and ~6.76 GBq/μmole for [18F]FLCM. Both radiotracers show favorable hydrophilic and low PPB profiles. In vivo PET scan and ex vivo study confirmed the brain uptake, renal and hepatic clearance, and minimal off-target uptake. The favorable pharmaceutical profiles establish a promising baseline, further rigorous in vitro receptor-affinity assays and dynamic brain imaging in an animal model are required to validate the translational diagnostic utility of these radiotracers.

Radiosynthesis and preclinical evaluation of 4-N-[¹⁸F]FDG-PBA as a novel PET tracer for tumor imaging

Conventional [¹⁸F]FDG exhibits inherent drawbacks in liver tumor imaging, especially high physiological uptake in normal hepatic tissues, which severely compromises the diagnostic accuracy of hepatic tumors. To address these limitations, we synthesized a novel tracer, 4-N-[¹⁸F]FDG-PBA, via conjugation of [¹⁸F]FDG...

[18F]FPyQCP: A Robust FAP-Targeted PET Tracer Integrating Covalent C−18F Labeling and Radiometal Chelation for Enhanced In Vivo Performance

Fibroblast activation protein (FAP) is a validated PET imaging target for the tumor microenvironment. Currently, most 18F-labeled FAP PET agents use Al−F chemistry, with Al18F-FAPI-74 leading clinically, whereas C−18F tracers perform less effectively. Here, we developed (4-quinolinoyl)glycyl-2-cyanopyrrolidine (QCP)-...

Preclinical Evaluation on the Radiohybrid Tandem [18F]La- and [225Ac]Ac-rhPSMA-10.1 for Targeted α-Therapy of Prostate Cancer.

Prostate-specific membrane antigen (PSMA)-targeted α-therapy holds promise for the treatment of prostate cancer but has increased side effects compared with therapy using β-emitting PSMA ligands. This emphasizes the need for precise imaging of the biodistribution of 225Ac-labeled ligands for patient selection and dosim...

2-Deoxy-2-[18F]Fluorosorbitol (18F-FDS) as a Potential PET Tracer for Renal Imaging and GFR Measurement: Comparative Assessment with 68Ga-EDTA and 99mTc-DTPA.

BACKGROUND Accurate assessment of glomerular filtration rate (GFR) is critical for monitoring renal function. This study aims to evaluate its feasibility as a PET radiopharmaceutical for renal imaging and GFR assessment. Its novelty lies in the direct, head-to-head comparison with 68Ga-EDTA and 99mTc-DTPA to comprehens...
