TL;DR

It is demonstrated that inherited disruption of the CTLA-4/LRBA axis profoundly remodels AIRRs, preferentially affecting CD8⁺ T-cell diversity while also constraining humoral immunity, and highlight a central role for this immune checkpoint pathway in maintaining adaptive immune repertoire diversity and clonal homeostasis.

PD-1 deficiency reshapes the TCRβ repertoire and T-cell developmental programs associated with susceptibility to lupus-like autoimmunity

The findings suggest that PD-1 deficiency is linked to disrupted TCRβ repertoire organization, extensive immune dysregulation, and an increased vulnerability to infection-induced lupus-like autoimmunity.

The CD160–HVEM axis at the crossroads of tolerance and immunity: lessons from cancer for autoimmunity and transplantation

Background The clinical success of immune checkpoint blockade (ICB) in oncology has validated the existence of molecular brakes that restrain T-cell immunity. While releasing these brakes unleashes antitumour responses, their physiological role is to maintain self-tolerance. This review explores the reciprocal paradigm...

Decoding autoimmune diseases through the adaptive immune receptor repertoires.

The adaptive immune receptor repertoire (AIRR), composed of T-cell receptors (TCRs) and B-cell receptors (BCRs), underpins antigen-specific immunity through extensive diversity generated by V(D)J recombination and junctional modification. Advances in high-throughput AIRR sequencing have enabled comprehensive characteri...

Soluble CTLA-4 as a context-dependent immune checkpoint: biology, biomarker potential, and therapeutic implications

Cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) exists not only as a membrane-bound immune checkpoint but also as a naturally secreted soluble isoform (sCTLA-4), generated primarily through exon 3 skipping. This review focuses specifically on splice-derived sCTLA-4 and examines its biological and clinical signific...

A four-domain framework for secondary immunodeficiency after CAR T-cell and T-cell-engaging antibody therapy

Chimeric antigen receptor (CAR) T-cell therapies and T-cell-engaging antibodies have transformed the treatment of hematologic malignancies but can induce prolonged, overlapping, and dynamically evolving immune deficits that are not adequately captured by hypogammaglobulinemia alone. Infection susceptibility reflects th...
