It was revealed that the self-assembled particles reversed SDS-induced gut dysbiosis, characterized by increased abundance of beneficial Dubosiella and decreased abundance of pathogenic Escherichia-Shigella and downregulated TFF1/NF-κB p65 protein expression.

Abstract

Background:Atractylodes lancea (AR) is a commonly used spleen-strengthening herb, but its clinical application is limited by the poor bioavailability of its active components. We found that AR spontaneously forms self-assembled particles during decoction, and bran-frying may further optimize this process to enhance its therapeutic efficacy against spleen deficiency syndrome (SDS). Objective: This study aimed to determine whether bran-frying enhances the efficacy of AR by forming more stable self-assembled particles with improved delivery performance, and to clarify the dual mechanisms by which these particles alleviate SDS through regulation of the TFF1/NF-κB pathway and modulation of the gut microbiota. Results: Compared with raw AR self-assembled particles (ARSs), bran-fried AR self-assembled particles (ARFs) exhibited higher atractylodin loading (1.17% vs. 0.96%), smaller particle sizes, and greater absolute zeta potential. ARFs also showed significantly higher cumulative release of all four active components, with atractylodin release approximately six-fold that of ARSs. Both particles significantly reduced intestinal inflammatory cytokine levels, restored barrier-associated molecular markers, and downregulated TFF1/NF-κB p65 protein expression. Via 16S rRNA sequencing, it was revealed that the self-assembled particles reversed SDS-induced gut dysbiosis, characterized by increased abundance of beneficial Dubosiella and decreased abundance of pathogenic Escherichia-Shigella. Fecal microbiota transplantation and antibiotic depletion experiments further verified the regulatory role of self-assembled particles on the gut microbiota. Conclusions: Both raw and bran-fried AR can form self-assembled particles, and bran-frying enhances their biological characteristics. The self-assembled particles exert spleen-strengthening effects through dual mechanisms involving regulation of the TFF1/NF-κB pathway and modulation of the gut microbiota.

Core-shell nanofiber-mediated delivery of fucoxanthin enhances its bioavailability and alleviates non-alcoholic fatty liver disease

Non-alcoholic fatty liver disease (NAFLD) remains a major health challenge, and although fucoxanthin (Fx) shows therapeutic potential, its poor stability and low bioavailability limit clinical application. In this study, a novel delivery system for Fx, a marine-derived xanthophyll carotenoid with anti-obesity, anti...

Next-Generation Solid Dispersion of Andrographolide Attenuated Inflammation and Oxidative Stress in an Activated RAW264.7 Macrophage-Based In Vitro Model Relevant to Ulcerative Colitis

Ulcerative colitis (UC) is fundamentally mediated by macrophage-driven immune dysregulation that amplifies mucosal inflammation. Andrographolide (AGD) has potent anti-inflammatory and antioxidant properties; however, AGD shows poor water solubility and bioavailability. To surmount these challenges, next-generation soli...

"Lego-Brick"-Style Supramolecular Self-Assembly for Naringin-Chlorogenic Acid-Glucosylrutin Carrier-Free Nanoparticles: Self-Assembly Mechanism, Release Behavior, and Anti-Inflammatory Activity.

Naringin (NG), a citrus flavonoid with anti-inflammatory properties, suffers from low solubility, limiting the full realization of its health benefits. Inspired by the modular assembly concept of "Lego-bricks", this study constructed naringin-chlorogenic acid-glucosylrutin carrier-free nanoparticles (NCGN) with high ap...

Development of self-assembled Arachis hypogaea L. skin polyphenol-wheat starch complex-based film for sustained release, antibacterial activity, and beef quality monitoring.

Although sustainable intelligent packaging is a critical frontier in food engineering, directly introducing natural bioactives tends to compromise film mechanical properties and hamper sustained release. Herein, an active and intelligent film was engineered via a multi-level supramolecular self-assembly strategy betwee...

Castanea henryi-resistant starch prepared with autoclave-assisted pullulanase treatment attenuated high-fat diet-induced obesity by regulating gut microbiota and metabolism.

The high glycemic index of Castanea henryi has significantly hampered its utilization for new product development. In this study, Castanea henryi starch was processed by autoclave-assisted pullulanase treatment (AAPT) to prepare resistant starch (RS). The structural property, crystal structure and thermal stability of...
