Abstract

Using KRAS and MEK inhibition, the authors demonstrate altered neoplastic cell fates and immune functions of cDCs, T cells, and macrophages. Addition of KRAS inhibition to PD-1 blockade enhances T cell– and macrophage-mediated antitumor immunity.

TNFR1 expression on cancer cells mediates immune escape and an immunosuppressive microenvironment

This study provides evidence for a key role of TNFR1, as expressed by malignant cells, in orchestrating a pro-tumor immunosuppressive environment through secondary inflammatory mediators. Results highlight the relevance of interfering TNFα/TNFR1 in cancer immunotherapy.

CD1d remodels the tumor-infiltrating myeloid populations controlling antitumor immunity

Evans et al. identify CD1d as a key regulator of myeloid cell diversity in breast cancer. CD1d targeting reshapes tumor immune infiltrates, suppresses tumor growth, and enhances immunotherapy efficacy, suggesting an actionable pathway for therapeutic intervention.

STAT3 inhibition for the induction of immunogenic cell death in multiple myeloma

ABSTRACT In a recent paper published in Blood Science, Sha et al. report that STAT3 inhibition eliminates multiple myeloma stem-like cells while eliciting hallmarks of immunogenic cell death. These highly intriguing findings connect stemness, integrated stress response, and antitumor immunity, yet in vivo validation re...

CCT2 Promotes Immune Escape in Colorectal Cancer by Regulating the JAK1‐STAT3‐PD‐L1 Axis

Colorectal cancer (CRC) is a major global health burden. While immune checkpoint inhibitors have greatly advanced cancer therapy, their therapeutic effects are unsatisfactory in microsatellite‐stable/proficient mismatch repair CRC. This study explored the molecular mechanisms underlying CRC immune evasion.

Statins suppress colon tumors by inhibiting KRAS condensates.

The unclear regulatory and transport mechanisms of Kirsten rat sarcoma viral oncogene homolog (KRAS) limit a comprehensive understanding of KRAS activity and the development of inhibitors. Wang et al. reported that farnesylation-induced KRAS phase separation drives colon cancer progression and resistance to G12C inhibi...

Foretinib triggers immunogenic cell death and enhances efficacy of anti-CD47 immunotherapy in colorectal cancer

Highlights • The dual-target c-Met/VEGFR kinase inhibitor foretinib reshapes tumor immune microenvironment (TIME) by inducing immunogenic cell death (ICD).• We identified an on-target mechanism of foretinib in transcriptionally activating Drath Receptor 5 (DR5), a key component of the extrinsic apoptotic pathway.• The...
