Biology

TL;DR

The ribosome–mRNA interface is identified as an intracellular determinant of antibacterial PNA susceptibility and rpsLI82N alters uS12-dependent 30S assembly and initiation-complex dynamics, thereby changing the effective occupancy of mRNA translation-initiation regions.

Design and applications of chimeric antisense oligonucleotides that bind TPP mRNAs as antibacterial agents.

The rapid emergence of antibiotic-resistant bacteria has created an urgent need for innovative therapeutic strategies that go beyond conventional small-molecule antibiotics. Among the most promising approaches is the use of nucleic acid-based therapeutics, particularly antisense oligonucleotides (ASOs), which offer hig...

A systematic identification of resistance determinants to antisense antibiotics suggests adaptation strategies dependent on the delivery peptide

Findings indicate that the specific identity of the CPP used plays a key role in determining its robustness against the evolution of resistance, and that laboratory evolution can illuminate the remaining gaps in the authors' knowledge on the mechanisms of action of asobiotics.

Tobramycin-Peptide Nucleic Acids Synergize with Polymyxin B and Other Bioactive Compounds to Inhibit Growth of Gram-Negative Bacteria

Resistance studies demonstrate that the TOB-PNA conjugate, both alone and in combination with PMB, maintains antibacterial activity over prolonged exposure, suggesting a higher barrier to resistance evolution than conventional TOB treatment.

Rescue of ribosomal protein bL27 in Streptococcus pneumoniae TIGR4 by an alternate protease

These results suggested the presence of an alternate bL27-processing protease in TIGR4 that was absent from D39, and shed light on an alternative mechanism for bL27 maturation, and indicate that some strains of S. pneumoniae harbor horizontally acquired redundant pathways for this essential ribosome processing step.

A Small Molecule Substrate Mimic to Inhibit Bacterial Hsp70 Chaperone Function.

Given the rapid spread of antibiotic resistance worldwide, it is imperative to develop new strategies against pathogenic bacteria. Here, we report a small-molecule inhibitor of bacterial heat shock protein 70 (Hsp70), called DnaK, a central component of the bacterial protein chaperone network. Screening of an in-house...
